Hi everyone, 

 

So I have DOR, and my mom had POF. Yesterday, I got to reading on Fragile X syndrome pre-mutation, and am starting to think I should probably be tested for this.

 

It's a genetic condition which, if you have the full mutation, can cause issues like intellectual disability, sometimes severe, especially in boys. If you only have the pre-mutation, it's usually asymptomatic other than POF. From my reading (reliable source), it seems that 12-14 % of women w POF who also have a relative with POF are affected by this pre-mutation. 

 

The reason this is relevant, other than explaining DOR/POF, is that with a pre-mutation, if I get pregnant with my own eggs, I would have an increased chance of having a child with the full mutation, aka intellectual disability. Knowing this would push me to go for donor eggs right away instead of trying one last time with my own.

 

Just wondering - any of you get tested for this? thoughts?

Waited to be financially and emotionally ready to consider treatments - 2017 dx DOR

 

1st IVF cycle June 2017

Microflare protocol with twice daily gonal-f 112.5, Menopur 75 and Suprefact 0.4

3 follies measurable on day 8 of STIMS.

Ovidrel trigger shot June 24

ER June 26

2 eggs retrieved, ICSI, both fertilized

1 embryo left at day 3 - Grade 2, 7 cells, 5%fragmentation. Assisted hatching, transferred June 29th

POAS 11dp3dt BFN, POAS 13dp3dt BFN, Beta July 13th BFN

 

RE estimates chances of success of repeat IVF w my eggs to be 15-20%

Waited on finances to recover for IVF#2, planned for Jan 2018

Estrogen priming protocol without OCP, estrace start Jan 13, Stim with Gonal-F 225 and Luveris 150/75 to start Jan 22

-- bust as estrogen did not suppress my natural ovulation, LH surge on day of baseline US pre-stims --Converted to natural cycle, but ovulated the morning of retrieval - converted to IUI

 

2nd IVF June 2018

estrogen priming mini-stim protocol without OCP (once daily Menopur 75 and Gonal-f 75, orgalutran)

also 6mo high dose Ubiquinol, avoided BPAs and had endometrial scratch)

6 follicules (!), 5 mature eggs retrieved, 3 fertilized with ICSI

1 day 5 grade 4AA blast transferred July 2 - BFP on FRER, strongest at 6dpt to 8dpt

Beta 7dp5dt 8.5, very low. FRER showing very pale 2nd line. Beta 10dp5dt 7.0. Beta 12dp5dt BFN

Hi Yogini,

 

I did get tested for this when I first got the DOR diagnosis at 34 and I was negative for it although I do have a family history of POF.  My grandma had 7 kids before the availability of BCPs which she went on at 33 and off at 39 and never had another period so it was assumed she went through early menopause.  Of the 3 girls she had my one aunt went through menopause in her early 40s but my other aunt and my own mother entered it in mid fifties, so looks like the bum gene was passed down to me but unrelated to fragile X.  Would have been nice to know all this family history before I started TTC...I would have made a point in starting earlier!  I think its totally reasonable to ask to be tested for this, especially if it would help in such important decision making.

I have DOR and got tested for Fragile X at 26, it came back negative for me. I also have no one in my family with fertility problems.


Me - 30 H - 32

TTC #1 since Dec 2013

October 2015 - AMH .43

 

IVF #1 - Jan 2016. 5 eggs retrieved, 4 matured, 0 fertilized.

IVF#2 - Jan 2016, Converted to IUI -BFN

IVF#3 - Feb 2017
10dp5dt - BFP Beta #1 55. 12dp5dt Beta  #2 132 Beta # 3 332 Beta #4 172,. Ectopic pregnancy in left tube. Pregnancy resolved itself.

FET #1 July 2017 - BFN

FET #2 August 17, 2017 - BFP Beta #1 10dp5dt - 800 , Beta #2 - 1501

08/08/2017 - Ultrasound 5 weeks 6 days shows 1 baby measuring right on track with heartbeat of 110 bpm!! yahoo.gif

14/08/2017 - Ultrasound measuring 6 weeks 6 days at 6 weeks 5 days, heartbeat of 138 BPM.

EDD - 04/05/2018 - Baby BOY! babyboy.gif

DS born May 7th, 2018.

 

Time for a sibling - No frosties, back to IVF

IVF #1 - 11 retrieved, 7 mature, 1 fertlized, did not make it to blast.

IVF #2 - 8 retrieved, 5 mature, 1 fertilized, 1 5 day blast frozen (5AB)

FET October 2019 - Natural FET cycle with trigger shot & HCG boosters. BFP HCG 10dp5dt - 316 12dp5dt - 687

Ultrasound at 6w1d shows perfect little baby, heart-rate 110BPM yahoo.gif

EDD - 22/06/2020 - Baby GIRL! babygirl.gif
